Police are searching a west London address after the arrest of two members of the Basque separatist group Eta, one of whom has been convicted for 22 killings, the Spanish interior ministry said on Friday.

Antonio Troitino, 55, and 39-year-old Ignacio Lerin Sanchez were arrested over alleged terrorist offences in Spain at around 4am on Friday, the Metropolitan police said.

Five suspected Eta members have been arrested this week in France, Spain and Britain as part of a joint crackdown on the group, the Spanish ministry announced.

Troitino, who was convicted of 22 murders in the 1980s, was freed in 2011 after serving 24 years. At the time, Spain's national court ruled that the six years Troitino had spent on remand could count as part of his sentence, which could not exceed more than 30 years under Spanish law at the time. The decision was reversed and an arrest warrant issued for Troitino.

Police in London said the two men had been arrested under Section 5 of the Extradition Act and had been taken to a central London police station. A 38-year-old woman had been detained at the same premises for alleged fraud offences, it added, while two other men had been arrested for alleged immigration offences. Arranz and Sanchez later appeared at an extradition hearing at Westminster magistrates court and were remanded in custody until 20 July.

Weakened by the arrests of most of its leaders and by waning support in the Basque region, Eta said in October it would give up its armed struggle, which lasted more than 40 years and claimed more than 800 lives.

But Madrid has declined to negotiate a definitive end to the conflict, insisting that Eta disband and hand over its weapons. Sixteen suspected Eta members have been arrested so far this year, mostly in France and Spain.

Blast hits Damascus as Turkey sends troops to border

BEIRUT/ISKENDERUN, Turkey (Reuters) - Rebel forces attacked Syria's main court in central Damascus on Thursday, state television said, while Turkey deployed troops and anti-aircraft rocket launchers to the Syrian border, building pressure on President Bashar al-Assad.

There was a loud explosion and a column of black smoke rose over Damascus, an Assad stronghold that until the last few days had seemed largely beyond the reach of rebels. State television described it as a "terrorist" blast.

Dozens of wrecked and burning cars were strewn over a car park used by lawyers and judges. State news agency SANA said three people were wounded by the bomb hidden in one of the cars.

The guerrilla attack in Damascus coincided with a Turkish military buildup on its border to the north and a growing sense of urgency in Western- and Arab-backed diplomatic efforts to forge a unity government and end 16 months of bloodshed.

Turkish military convoys moved slowly towards the Syrian frontier, reacting to Syria's shooting down of a Turkish warplane over the Mediterranean on Friday. A Turkish official said they were reinforcing air defenses.

Prime Minister Tayyip Erdogan, speaking after Friday's incident, ordered his troops to treat any Syrian military element approaching the border as a military target.

He gave no details, but this could cover Syrian forces pursuing rebels towards the border, or patrolling helicopters or warplanes. Syria said at the weekend that it had killed several "terrorists" infiltrating from Turkey.

A first substantial convoy of about 30 military vehicles, including trucks loaded with anti-aircraft missile batteries dispatched from Turkey's coastal town of Iskenderun, was moving slowly towards the Syrian border 50 km (30 miles) away.

A Reuters reporter near the town of Antakya saw the convoy moving out of the hills and through small towns on a narrow highway escorted by police.

Early on Thursday, another convoy left a base at Gaziantep and headed for Kilis province, the site of a large camp for Syrian refugees. Video from the DHA agency showed the convoy, of about 12 trucks and transporters, filing through the gates of the base past the hanging Turkish red flag with white crescent moon and star.

David Hartwell, Middle East analyst at IHS Jane's called the Turkish action a 'pragmatic, rational response' after the shooting down of the Turkish aircraft, that Syria insists was flying low and fast in Syrian air space. "Damascus has been warned once. I doubt there will be a second warning."

Turkey, in the front line of Western efforts to press Assad from power, hosts over 33,000 Syrian refugees on its southeastern border as well as units of the rebel Free Syria Army (FSA).

REGIONAL FEARS

"I can confirm there are troops being deployed along the border in Hatay province. Turkey is taking precautions after its jet was shot down," a Turkish official said on condition of anonymity.

He said he did not know how many troops or vehicles were being moved but they were being stationed in the Yayladagi, Altinozu and Reyhanli border areas. He said anti-aircraft guns were being stationed along the border.

At a fortified Turkish border outpost near Reyhanli, a low building with red tile roof perched on a hilltop, troops stood on an observation platform looking across into Syria; but there was no sign of any unusual activity.

Members of the FSA, talking close to the border, told Reuters they did not believe the Turkish deployments were on a large scale or aimed at any crossborder intervention.

"The Turks know that any large scale military action would need international support," a senior FSA commander, who spoke on condition of anonymity, said.

Turkey has in the past spoken of opening a humanitarian corridor on Syrian soil, if the refugee flow grew unmanageable or if the violence and killing became intolerable.

Wary of igniting a regional sectarian conflagration, it has always insisted this would be possible only with United Nations backing. Western- and Arab-backed efforts to forge a joint diplomatic approach with Russia have so far failed.

The FSA has stepped up pressure on Damascus in recent weeks, Thursday's attack marking a new stage in the campaign.

On Wednesday, rebels stormed a pro-Assad Syrian television channel and militants have targeted police and security personnel barracks. In April militants fired rocket-propelled grenades at the Central Bank building.

A senior opposition official said Syrian opposition groups would reject a political transition plan proposed by peace envoy Kofi Annan unless it explicitly required Assad to step down before a unity government is formed.

Diplomatic sources at the United Nations said Annan's proposal did not stipulate Assad's resignation although it does say the unity government could not include figures who jeopardize stability.

"The proposal is still murky to us but I can tell you that if it does not clearly state that Assad must step down, it will be unacceptable to us," said Samir Nashar, an executive member of the international Syrian National Council.

DIPLOMATIC STALEMATE

Annan's transition proposal is one of the main topics that Russia, the other four permanent U.N. Security Council members and key players in the Middle East will discuss at a meeting in Geneva on Saturday, according to United Nations diplomats.

Rebel fighters locked in the war to topple Assad said there was no part of the plan they could accept, and they had lost patience with U.N. envoy Annan's peace-making efforts.

"This is just a new labyrinth. It is new silliness for us to get lost in and haggle over who can participate and who can't," said Ahmed, a Free Syrian Army (FSA) fighter in Homs, epicenter of the revolt against four decades of Assad family rule in which the more than 10,000 people have been killed, by a U.N. count.

A member of the rebel group in Damascus suburb was also dismissive.

"I'll be direct. The FSA is doing its work, and it is not looking to the outside world. We don't want a transitional government unless it is the one formed by rebel military councils. The world is conspiring against the Syrian revolution," he said.

In April, Annan tried to implement a ceasefire to quell violence before embarking on peace talks. But the truce failed to take hold.

VIRGINIA ANNOUNCES CERTIFIED FARM SEEKER PROGRAM

In a reversal of trends over the past couple of generations, young people today seem to be returning to the farm, or they would if they could just find one. Farming is an occupation that is asset heavy ? land, equipment, buildings, machinery, livestock ? and all of those assets are expensive. A mid-size farm, if you can find one for sale, can run into hundreds of thousands or even millions of dollars. Some farmers are making a living on thirty acres or fewer. But if a would-be farmer really wants a farm with all the acreage, barns, sheds, tractors, planters, sprayers, combines and harvesters, what is he or she to do?

Thanks to a new program offered by the Virginia Department of Agriculture and Consumer Services (VDACS) and the Virginia Farm Bureau?s Young Farmers Committee, would-be farmers can become a Certified Farm Seeker, a designation that lets landowners know you are serious about farming.

The Certified Farm Seeker (CFS) program is designed for farmers at all levels: beginning, established or transitioning. The program?s goal is to provide individuals who want to farm with the tools needed to successfully demonstrate their farming commitment and vision to interested landowners. Those who earn the title Certified Farm Seeker stand out from the crowd. Landowners know these individuals are serious about establishing a farm enterprise and have completed the planning necessary for long-term success.

Agriculture is a business; in fact, it is Virginia?s largest industry. And like any other business, would-be farmers need a sound business plan as a first step on the road to their goal of farm ownership. The CFS Program is designed around helping interested farmers produce a business plan and resume, as well as demonstrate on-farm experience. Five Whole Farm Planning Modules are available as resources to guide seekers through this process. Each module contains objectives, questions and possible activities to help beginning farmers reach their goals. The curriculum is designed to be flexible and can be applied to farmers at every experience level.

When aspiring farmers complete the CFS program, they gain many rewards. They receive professional review of their business plan at reduced or no cost. They receive priority in the Virginia Farm Link database, an online resource designed to link farm owners interested in exiting agriculture with those seeking farms and farm businesses, thereby increasing the farm seekers? chances of being contacted by a landowner. They receive invitations to numerous networking and social opportunities to interact with other farmers and landowners. And they may even receive time with a transition mediator or attorney at reduced or no cost.

?I was one of the lucky ones,? said Matthew J. Lohr, VDACS Commissioner. ?I grew up on a family farm and retain an interest in that farm today. But not everyone is so lucky, and for those who want to turn their childhood fascination with machinery, plant and animals into a viable business, the Certified Farm Seeker Program is just the tool to get them started and to ensure their success.?

Historic handshake: Queen meets ex-IRA chief

Martin McGuinness, a former commander of the Irish Republican Army met with Queen Elizabeth in Northern Ireland. It was a historic moment decades after the IRA led a bloody fight against British rule. NBC's Michelle Kosinski reports.

Thirty-five years ago, Queen Elizabeth's silver jubilee was greeted with graffiti declaring "Victory to the IRA, stuff the jubilee."

Wednesday marked a highly significant turnaround as the queen, in her diamond jubilee year, met and shook hands with a?onetime senior Irish Republican Army commander who once stood against everything she represented and even considered her a legitimate target.

As a British person and a journalist, I never thought I'd see this day.


This is because I grew up with Northern Ireland. What does this mean? It means watching with incredulity as the IRA targeted the British establishment, including a sitting prime minister -- almost succeeding in assassinating her.

The "troubles," as they were diplomatically called, became part of everyday life. We watched clashes with soldiers on television. News of bombings was a constant drip-drip in the news. It was one of those things that as a boy and a young man, I thought would never end.?

So the meeting with Martin McGuinness, the first between the queen and a senior member of the IRA or its political wing Sinn Fein, is a landmark in the peace process 14 years after the militant group ended its 30-year campaign against British rule.

On Tuesday, she held a private meeting with relatives of the 11 people killed in a 1987 bombing in Enniskillen, an?attack that sparked a wave of revulsion against the IRA and helped convince its leadership to engage in the peace process.

Few will know what the queen was thinking. But surely it was a difficult event for her, and not just because of the more than 3,500 killed in the conflict, 1,800 of whom were innocent civilians, according to The Guardian.

It is doubtless especially poignant for her because her cousin, Lord Mountbatten, was killed in 1979 when the IRA?blew up his boat in Southern Ireland.?He was the man who many believe was responsible for the queen's marriage to Prince Philip, and was a guiding influence to the heir-to-the-throne Prince Charles.

The countless threats her family has lived with must have contributed to a feeling of unease ahead of the meeting. But the queen's real thoughts will probably never be known, nor will her reaction when she was advised to perform this duty.

And it wasn't only the queen who was taking a chance -- it came at a cost for McGuinness too. He was being branded a traitor, with a lot of republicans saying that he has sold out and betrayed the principles they stand for.

Republicans protested against Wednesday's meeting, and McGuinness' decision could hurt his political ambitions.

British reaction on a political level has by and large been supportive, despite the bitterness and painful memories of the past.

Norman Tebbit, a former Conservative Party Chairman and a survivor of a deadly bombing in the seaside city of Brighton that targeted Prime Minister Margaret Thatcher, saw it as a victory over the IRA.

Before Wednesday's event, Tebbit wrote in?The Telegraph?that the meeting would be a victory for the queen, the monarchy and Great Britain:?

"I am glad that Mr McGuinness appears to have now accepted on behalf of IRA/Sinn Fein the sovereignty of Her Majesty over Northern Ireland, and I hope that this is a step towards a public recompense and confession of his regret for the violence unleashed by them in his name."

While the meeting does not mark the end of tensions in Northern Ireland, it draws a line under a conflict that cost the lives of thousands and beset the queen for half of her reign.

(AP) ? A North Korean woman said Thursday that she was tricked into defecting six years ago by South Korean agents who offered to arrange a reunion with her father who went to the South during the Korean War.

The rare public account that 66-year-old Pak Jong Suk told to local and foreign reporters at a news conference at the People's Palace of Culture in Pyongyang could not be independently confirmed. South Korea's Unification Ministry said it was investigating and would release its findings later. It didn't elaborate.

"I am an ingrate who had betrayed my motherland to seek better living while others devoted themselves to building a thriving nation, tightening their belts," said Pak, clad in a pink traditional Korean dress.

It is unusual for North Korea to hold and televise a news conference for foreign as well as local media featuring ordinary citizens ? particularly a former defector. It was not possible to immediately verify whether Pak spoke on government orders or of her own volition, but North Korea has bristled in recent weeks at allegations of rights abuses and maltreatment of repatriated defectors.

Pak said she slipped undetected across the Tumen River from the North Korean city of Chongjin into China in March 2006, after being promised that she would be reunited with her father in the Chinese city of Qingdao. She said she hoped to get money from him. Three months later, after paying smugglers, she said she was tricked by South Korean intelligence agents into boarding a boat that landed in South Korea.

Pak said her father was unconscious because of brain surgery and did not speak to her before he died, two months after her arrival. She lived in South Korea before returning to North Korea by plane on May 25 this year because she had become disillusioned with life in the South. She said defectors are paid by South Koreans to slander North Korea.

The circumstances of how she returned to the North were not clear. She said she lives now in Pyongyang with her son, a teacher, and his wife, who appeared at her side at the news conference, parts of which were later broadcast on state television.

"When I deplaned, quieting my thumping heart, I was stunned by the cordial reception," Pak said, revealing her surprise that she wasn't treated more harshly upon her return.

The U.S. State Department said in its annual human rights report last month that the relatives of defectors face "collective punishment" if a family member defects, and that defectors face harsh punishment if repatriated to North Korea.

Pyongyang denies the allegations of human rights abuses.

The Korean Peninsula has remained divided by a heavily fortified border since the Korean War ended in a truce in 1953. Since then, more than 23,000 North Koreans have defected to South Korea, mostly by crossing into China and then making their way to a third country before being sent to the South, according to the South Korean government.

North Koreans automatically are granted South Korean citizenship under the South's constitution, but undergo rigorous questioning after arriving in the country.

"I deserve punishment. But Kim Jong Un did not blame me but was so kind as to enable me to enjoy the greatest happiness," Pak said at the news conference.

Kim Jong Un became North Korean leader after the December death of his father, Kim Jong Il.

The transition of leadership to Kim is taking place amid concerns about North Korea's ability to feed its 24 million people. The United Nations said in a recent update on the North's humanitarian situation that the food supply remains tenuous for two-thirds of the population, and many North Korean children are not getting the food, medicine or health care they need to develop physically or mentally.

In the year 2007 was when the first Trojan surfaced according to PC World.? The malware, which infects both Windows and Mac computers, creates a botnet by changing the machines accessed domain name server (dns).

A Domain Name Service is how the web organizes its addresses.? It is a system that lets you simply type in ?mashable.com? instead of some kind of long IP address filled with letters, decimals and numbers.? Your computer talks to a DNS server operated by your Internet Service Provider (ISP) to find all the websites that you visit every day.

The trojan, called a DNS Changer, redirects your computer?s DNS queries from your ISP?s server to one created by the trojan?s creators which hijacks all Internet traffic from your machine.? That way, the bad guys can send you to hacker-created websites filled with ads whenever they want.

There is good news!? The FBI shut down the operation, called Rove Digital, last November when they arrested six Estonian nationals behind the botnet and shut down their malicious servers to ensure infected computers wouldn?t be cut off from the Internet entirely, the FBI set up its own DNS servers.

But, of course there is some bad news.? Those servers will soon be shut down. They were originally going to run only four months, but a judge ordered an extension of their operation until July 9 since it?s estimated that hundreds of thousands of computers are still infected.

If a machine still has the trojan and tries to access the web on July 9, it won?t be able to access anything.? With Internet access cut off, it would be very inconvenient to download and install anti-virus software.

Cornerback Alphonso Smith doesn?t care whether the Lions are trying to trade him, and he won?t care until they do.

Smith, picked by the Broncos in the second round of the 2009 draft after Denver shipped a 2010 first-rounder to Seattle, was sent to Detroit after only one season for tight end Dan Gronkowski.? Earlier this year, rumors surfaced that the Lions could move Smith in an effort to beef up their secondary.

?Please forgive my frankness,? Smith told Dave Dye of FOXSportsDetroit.com.? ?To be blunt with you, I really didn?t care.? It didn?t happen.? It still may happen.? You never know.

?I don?t care about reports or rumors.? Only thing I can focus on is I?m a Detroit Lion and I?m a Detroit Lion until I?m not.?

His preference would be to continue to wear Honolulu blue.

?I want to be here,? Smith said.? ?I?ve grown a lot with the scheme and the players and the things that the coaches want.? But that decision is way above my pay grade.? My job is to come in and play, not deal in personnel or whether I?m getting traded or not. . . .? Really don?t care.?

Smith does care that he?s in a contract year.? With his first-ever shot at free agency coming in March, he has every reason to play well, regardless of where he ends up playing.

If he stays in Detroit, he may have a chance to play plenty.? He could end up filling the vacancy in the starting lineup created by the departure of Eric Wright via free agency.

?One of the ways I try to separate myself is making plays,? Smith said.? ?As I look around, if you talk about playmakers in the secondary, I?ll probably be the first on that list.? That?s how I separate myself. . . .? Do my job.? Don?t give up any big ones and make plays when it?s there.?

Regardless of whether Smith gets traded, the Lions need someone to not ?give up any big ones? and ?make plays when it?s there? in 2012.? If not, the Lions won?t get any farther than they did in 2011.

CompTIA is a non-profit organization that specializes in validating knowledge of a wide range of computing professionals. They offer tests in many different vendor neutral areas. These include computer technology skills, security, networking, server administration and training. The test that CompTIA administers is designed to validate basic networking knowledge and skills. It is questions long and you have 90 minutes to complete it. The test questions are multiple choice and you need a score of 720 to pass. That passing score shows you understand network technology, network media, topographies, network devices, network management, tools and network security.

"Safety" can't sustain its own offbeat energy. By the end, even Aubrey Plaza would roll her eyes at it.

Neatly, the script embarks on one journey while dangling the possibility of another: the prospect of taking a sudden leap from comic reality into the realm of pure imagination.

"Safety Not Guaranteed" is most vibrant and vital at its edges, in the way that the characters interact with each other while waiting for something to happen.

It's brisk and assured and never begs the audience's indulgence. No time is wasted. The movie is, at every moment, either funny or pushing the story forward, or both.

The film is modest but skillful and heartfelt, spiced just so by Plaza and company.

Safety Not Guaranteed casts an enchanting spell from its opening scene.

It is an offbeat charmer that is fun and engaging from a storytelling standpoint, but one that also brings a surprisingly rich emotional payoff.

Unlike time travel or jet-setting, "Safety Not Guaranteed" shows that making movies can still be more about the journey than the destination. Whether or not you have first-class tickets.

"Safety Not Guaranteed" is eccentric enough to get mistaken for an uplifting fantasy, but it's Plaza who belongs in the penthouse.

Hope is a powerful thing and Safety Not Guaranteed delivers in its tale of longing, deliverance and connection during the here and now.

An intriguing yet erratic effort...

Less of a philosophical argument and more of a character piece driven deep into the heart by Duplass and Plaza.

Have you ever wondered what mumblecore sci-fi would look like? Wonder no more.

Rather than trying to beat Hollywood at its own game of high-tech gadgets and weaponry, director Colin Trevorrow and writer Derek Connolly achieve a sly mix of the insane and the mundane.

A character-driven piece about regret and true partnership - our basic, primal need for someone to take the journey with us even if safety is not guaranteed.

Aubrey Plaza is the best thing about this iffy mock-sci-fi doohickey.

As a hipster rom-com about people trying to shake off their pasts to make present-day connections, it's pretty satisfying.

A strange, light-hearted bit of quasi-sci-fi, with no small amount of heart.

The material is played mostly for laughs and succeeds in that regard. The undercurrent of lament in Safety Not Guaranteed, though, is what holds the film together.

Blood and excrement stained a set of wooden boards, fitted with hinges so they could be formed into a fighting pit.

Six dogs, heavy chains running from their collars to the unplanted dirt, were pinned to the ground behind a bizarre high desert house that police say was a center for the illegal blood sport of dogfighting.

?It smelled like the worst kennel you?ve ever been in in your life,? said Los Angeles Sheriff Lt. Larry Gregg, who described the scene for NBCLosAngeles.com.

Deputies raided the house in the community of Littlerock Saturday night, arresting twelve people and removing eight dogs, Gregg said.

Humans and animals were crowded into a bizarre backyard compound made entirely of old garage doors, Gregg said. Among them was a man the Humane Society of the United States says is a suspected kingpin of Southern California dogfighting.

?You walk into a garage where it looked like they were doing the fighting, and there were these two-by-fours hinged together,? Gregg said. ?There was blood all over the boards, and fecal matter."

Dogfighting, which dates from the 1800s, is illegal in most countries, but continues as an underworld activity.

Participants pay entrance fees and place bets on which dog will win the fight. Owners of winning dogs can charge stud fees for breeding more fighters.

The dog of choice for these fights in the U.S. is the American Pit Bull Terrier, bred for fighting in the early 1800s from English Bulldogs and a terrier breed.

??Dogs are pitted against each other in fights that end with the death of one or both dogs,? Goodwin said. ?These animals suffer and die just for the entertainment and gambling desires of the people who are there.?

Dogfighting has been on the wane in the United States since football player Michael Vick pleaded guilty in a high-profile case in 2007, drawing attention to remaining fighting and gambling rings, Goodwin said.

Since the Vick case, incidents of dogfighting have dropped in half in the U.S., he said, largely due to an increase in police enforcement.

The tip that led sheriff deputies to the Littlerock house came from a hotline set up by the Humane Society aimed at tracking down dogfighting rings. The animal rights agency has a dogfighting fact sheet online, and its tipline is 1-877-NO2FITE.

But Goodwin said the fights continue in many places, including Southern California.

SANAA (Reuters) - A road linking Yemen's capital to an oil-producing province was opened for the first time in more than a year on Sunday, after the army and tribal fighters agreed to withdraw from positions along the route, military officials said.

Yemen's Republican Guard had skirmished with tribal groups in the area, blocking deliveries of gas and other products from Maarib to the capital.

Both sides agreed to pull out after negotiations, said an official from Yemen's military committee - a body set up to separate belligerent factions of Yemen's army and tribal fighters following months of unrest in the impoverished country.

The tribal groups backed an uprising which started last year and eventually ousted Yemeni president Ali Abdullah Saleh. The Republican Guard is led by Saleh's son.

Their clashes have added to the insecurity in a country which has also seen repeated bombings of the Maarib oil pipeline, which feeds Yemen's main oil refinery and remains inoperative.

Saleh gave way to his deputy, Abd-Rabbu Mansour Hadi, in February under the terms of a power transfer deal brokered by Saudi Arabia and endorsed by Washington, both alarmed by the rise of al Qaeda-linked Islamists in Yemen amid the political upheaval.

Washington, which wants Hadi to unify the military and turn it against al Qaeda, backed a military offensive against Islamist strongholds in southern Yemen that began last month.

The military says it has driven Islamist fighters from towns the militants seized in early 2011. Yemen's al Qaeda branch claimed responsibility for the killing of the top military commander in the region last week.

Stopping and starting cancer cell cycle weakens and defeats multiple myeloma

Weill Cornell Medical College researchers have devised an innovative boxer-like strategy, based on the serial use of two anti-cancer drugs, to deliver a one-two punch to first weaken the defenses of multiple myeloma and then deliver the final knock-out punch to win the fight.

The study, published online by the journal Blood, is the first to show that precise timing of therapies that target a cancer cell's cycle ? the life phases leading to its division and replication ? disables key survival genes, resulting in cell death. The drug that delivers the weakening jab at the cell cycle is the experimental agent PD 0332991, which allows bortezomib, a proteasome inhibitor already approved for use in myeloma and lymphoma, to land the final defeating blow at lower than normal doses.

While this is potentially good news for patients with multiple myeloma, a cancer of blood plasma cells that is currently incurable, the study suggests that using this therapeutic strategy could also work for other tumor types, says the study's senior investigator, Dr. Selina Chen-Kiang, professor of Pathology and Laboratory Medicine and of Microbiology and Immunology at Weill Cornell Medical College.

"Because robust functioning of the cell cycle is crucial to cancer growth and survival, this mechanism-based strategy could theoretically be used against many kinds of cancers," she says.

"Based on the genetics of a patient's tumor, we could pair PD 0332991 with the right cytotoxic partner drug to both inhibit cancer cell division and sensitize the cells for that knock-out punch," says Dr. Chen-Kiang. "We are very excited about the promise of this approach."

In fact, physicians at Weill Cornell have opened two new human clinical trials, one in multiple myeloma and one in mantle cell lymphoma, based on the findings of this study in a mouse model as well as on a previous phase I clinical trial led by Weill Cornell investigators that tested PD 0332991 in patients with mantle cell lymphoma.

Playing Havoc with the Cancer Cell Cycle

Dr. Chen-Kiang and her laboratory colleagues have long studied genes and proteins that control the cell cycle and cell suicide (apoptosis) in cancer. Cancer is fundamentally a disease of uncontrolled cell proliferation, where cells are able to continuously divide. In contrast, cell division in a healthy individual is regulated by the cell cycle, an orderly sequence of programmed gene expression in which the cell is driven through various checkpoints by a highly regulated network of proteins.

Cyclin-dependent kinases (CDKs) are molecules that power the progression of the cell cycle through its four phases. For example, CDK4 and CDK6 help move cells through the first G1 "gap" phase to later phases where the cell splits in two. In many cancers, these two enzymes are over-expressed, ensuring continual growth. Therefore, targeting CDK4 and CDK6 to shut them down has long been a goal of cancer drug discovery, but clinical success, so far, has been disappointing because of lack of effectiveness as well as drug toxicity, says Dr. Chen-Kiang.

PD 0332991, a small molecule synthesized by Pfizer, is different because it is exceptionally selective for CDK4 and CDK6, she says. The drug initially did not receive much attention because it is also reversible, meaning that it needs to be used continuously to inhibit CDK4 and CDK6; withdrawing it would reactivate these enzymes, stimulating growth.

But Dr. Chen-Kiang had been searching for a drug that she could use for her selective cell cycle-based therapy ? the idea being that playing havoc with a cancer cell's cycle would fatally weaken it when more traditional anti-cancer drugs are used sequentially.

"Given that the gene expression program is coupled to the cell cycle, we hypothesize that inhibition of CDK4/CDK6 maintains gene expression programmed for early G1, while preventing the expression of genes scheduled for other cell cycle phases," she explains. "And because metabolic needs in tumor cells differ from normal cells, this prolonged arrest in G1 would create an imbalance in gene expression that preferentially sensitizes tumor cells to cytotoxic drugs, allowing for low-dosage treatments."

"Because PD 0332991 is also reversible, we further hypothesize that release from G1 by removal of the inhibitor would synchronize the cell cycles, but may not synchronize gene expression schedules," Dr. Chen-Kiang adds. "This tension between cell cycle synchronization and differential gene expression synchronization further weakens the tumor cells during their progression, as does the heightened metabolic load and demand for energy to replicate DNA."

Loss of Survival Protein

This is exactly what the researchers found. By using PD 0332991 several times to induce a prolonged arrest of G1, and then a release from that arrest, the cells were sensitized to killing by bortezomib. They found this to be the case in laboratory studies of primary myeloma tumor cells and in mice, which were left with healthy bone marrow cells.

"We found bortezomib, even when used in a low dose, was significantly more effective when the cancer cells were sensitized by our strategy," says the study's first author, Dr. Xiangao Huang, assistant research professor of Pathology and Laboratory Medicine at Weill Cornell Medical College.

In exploring the underpinning mechanism, the researchers discovered that prolonged arrest in G1 markedly enhances cell suicide, induced by bortezomib. They discovered that during this phase, the cell loses a protein called IRF4, an essential survival factor for myeloma cells, and gains several pro-apoptotic proteins.

"These findings demonstrate for the first time that key survival and apoptotic genes are regulated by the cell cycle in cancer cells, and suggest new molecular targets for intervention," Dr. Chen-Kiang says.

"This work represents the seamless integration of basic biological research on the cell cycle and direct medical application in clinical trials," she adds. "Both the tools available to us, and our unique location at NewYork-Presbyterian/Weill Cornell Medical Center, allow us to move biological research forward while rapidly translating our findings to therapy."
