This is a great conversation I had with a client yesterday.

When we first started working together more than a year ago, he said that he suffered from low self-esteem and that this was one of the issues he wanted to work on.? Unsurprisingly, this lack of self-esteem turned out to be related to bad/abusive relationship experiences he'd had, and we've healed a lot of that.? Still, I thought it was time to ask him how he was doing, overall, with the question of self esteem.? I believe that it's important to check in periodically and see how an issue is improving--or not improving--globally.? This is how I can assess if there is something that needs to change in my own approach--do we need to do some EMDR around past negative events, do we need to go into a more somatic process, is it a spiritual/meaning of life/existential issue, etc.

So, he said something very interesting:? "My self esteem still isn't that great but my self worth has come up a lot."

"That's great.? What would you say the difference is?"? I asked, intrigued.? This appears to me to be a very sophisticated and accurate way to talk about it.? This client is very intelligent (far more than he used to think he is, so coming to an awareness of that has been really helpful for his self worth, for sure.)

He said, "Self esteem...it changes a lot depending on who I'm around, how I look that day, what other people might think of me.? Self worth doesn't change that much. I know now that I'm a good person and deserve more from my relationships than I used to think."

I thought this was great.? Self worth is like the core; a sense of being a deserving human who rightly occupies a place in the world, gives and receives love, is treated and treats others with dignity and respect, is effective and capable, competent and strong.? It's the core.? Self esteem, well, it changes; it might be better on the days you have on an awesome pair of jeans, or a good hair day, or you get a smile from a stranger.??

So yeah.? I think the core of good healing is the self worth. I would like to see self esteem come up too, but if the self worth is gaining ground and growing, that's the most important thing and likely a lot of the self esteem stuff will follow it.? Incidentally, how awesome is it for a man to openly say that this is part of what he wants for himself out of healing?? I told him that one of the reasons people love him (because they really do) is that he's brave.? He gets out there and deals with himself and thinks about things, and people admire that even though it scares the pants off them initially.? I think it's hard for a lot of men to even admit they have self-esteem issues much less get assistance with them.? It takes a courageous guy to face that one down and work with it.? She has worked with him since kindergarten. We have additionally spent many, many hours supporting his reading and writing skills. We are fortunate ? Craig has a Bachelor of Arts Degree in Education (and another in Music) so he is very well prepared to support H as his home facilitator. And me, well? I am a special education teacher, with a focus in supporting educators in understanding the experience of students with autism. Needless to say ? it would not be an understatement to suggest that we are, indeed, prepared to oversee H?s education.

I realize that distance education is not the answer for every child. Like so many of the aspects of parenting a child with autism? ? a decision like this affects the family at the systems level. We have arranged our lives so that this works for us. It is a match for H and we are fortunate that we have been able to shift our lives so that this choice is feasible.

I guess the point here is that we have been able to find a way to make things work for our child and our family. There is not one right way? or a single correct path. Our lives may not be what we had envisioned when we set out on this journey?

They are, in fact, richer and much more interesting than we had ever imagined, and our child is learning almost as much as we are. No. Really!!

Protein-labeling technique allows high-resolution visualization of molecules inside cells

CAMBRIDGE, Mass. -- The glowing green molecule known as green fluorescent protein (GFP) has revolutionized molecular biology. When GFP is attached to a particular protein inside a cell, scientists can easily identify and locate it using fluorescence microscopy. However, GFP can't be used with electron microscopy, which offers much higher resolution than fluorescence microscopy.

Chemists from MIT have now designed a GFP equivalent for electron microscopy a tag that allows scientists to label and visualize proteins with unprecedented clarity.

"With things that may appear only a few pixels across by fluorescence microscopy for example, a mitochondrion you can't make out any of the internal features. But with electron microscopy it's very easy to discern the intricate internal structures," says Jeff Martell, a graduate student in chemistry at MIT and lead author of a paper describing the new tag in the Oct. 21 online edition of Nature Biotechnology.

The new tag could help scientists pinpoint the locations of many cell proteins, providing new insight into those proteins' functions, according to the researchers.

Improving on nature

Dubbed APEX, the new tag is similar to naturally occurring proteins that have been tried as imaging labels for electron microscopy. Horseradish peroxidase (HRP) is one commonly used tag, but it works only in a few compartments of a cell. Other recently developed tags work throughout a cell but are technically challenging to use because they require light to be shined on the sample and oxygen to be bubbled through it.

To improve on these methods, the researchers started with a protein similar to HRP, called ascorbate peroxidase (APX). APX is more versatile than HRP because it can function within a cell's cytosol, in the main cavity of a cell.

Both HRP and APX belong to a class of enzymes called peroxidases, which remove an electron and a proton from other molecules in a process known as oxidation. Every peroxidase has different targets, and one of HRP's main targets is a molecule called DAB, which when oxidized can be visualized with electron microscopy. The researchers genetically engineered APX so that it would also target DAB.

To use this new APEX tag (for "engineered APX"), the researchers deliver, into a living cell, a small ring of DNA containing the APEX gene joined to the gene for the protein they plan to image. The cell then produces the target protein, bound to the APEX protein.

Next, the researchers need to deliver DAB, which is not normally found in cells. This delivery takes place during the process of "fixing," or stabilizing cells, which must be done before they can be imaged with electron microscopy.

When the APEX protein oxidizes DAB, it generates radicals that rapidly clump together into a tarlike polymer. That polymer can be detected through electron microscopy, allowing the researchers to pinpoint the location of the target protein.

A biological question resolved

To demonstrate the usefulness of their new tag, the researchers set out to resolve an open question regarding the location of a calcium channel protein discovered last year. Two research groups identified the protein and reported that it is located within mitochondria, but they had conflicting theories as to its precise location and orientation. Using the new imaging technique, the MIT-led team labeled the protein and determined that it is embedded in the inner mitochondrial membrane and faces into the innermost part of mitochondria, the mitochondrial matrix.

The team also showed that the new tag can label proteins throughout the cell not only within mitochondria but also in the nucleus, the endoplasmic reticulum and the cytosol.

Martell and Alice Ting, the Ellen Swallow Richards Associate Professor of Chemistry at MIT and senior author of the Nature Biotechnology paper, invented the new technology. How a fish 'broke' a law of physics

ScienceDaily (Oct. 21, 2012) ? Silvery fish such as herring, sardine and sprat are breaking a basic law of physics, according to new research from the University of Bristol published October 21 in Nature Photonics.

Reflective surfaces polarize light, a phenomenon that fishermen or photographers overcome by using polarizing sunglasses or polarizing filters to cut our reflective glare. However, PhD student Tom Jordan and his supervisors Professor Julian Partridge and Dr Nicholas Roberts in Bristol's School of Biological Sciences found that these silvery fish have overcome this basic law of reflection -- an adaptation that may help them evade predators.

Previously, it was thought that the fish's skin -- which contains "multilayer" arrangements of reflective guanine crystals -- would fully polarize light when reflected. As the light becomes polarized, there should be a drop in reflectivity.

The Bristol researchers found that the skin of sardines and herring contain not one but two types of guanine crystal -- each with different optical properties. By mixing these two types, the fish's skin doesn't polarize the reflected light and maintains its high reflectivity.

Dr Roberts said: "We believe these species of fish have evolved this particular multilayer structure to help conceal them from predators, such as dolphin and tuna. These fish have found a way to maximize their reflectivity over all angles they are viewed from. This helps the fish best match the light environment of the open ocean, making them less likely to be seen."

As a result of this ability, the skin of silvery fish could hold the key to better optical devices. Tom Jordan said: "Many modern day optical devices such as LED lights and low loss optical fibres use these non-polarizing types of reflectors to improve efficiency. However, these human-made reflectors currently require the use of materials with specific optical properties that are not always ideal. The mechanism that has evolved in fish overcomes this current design limitation and provides a new way to manufacture these non-polarizing reflectors."

Surface tablet pre-ordering starts now at $499; ships Oct. 26

Microsoft's Surface RT, the company's surprise entrance into the tablet world, will start at $499 when it ships on Oct. 26. Pre-ordering starts today?at Surface.com. More importantly, I got a chance to play with some of them on Monday, and my impressions are below.

The base-price tablet has 32GB of internal storage (twice what the $499 iPad has, for those keeping track). For $599, you can buy that tablet with a Touch Cover keyboard add-on, and for $699, you get a 64GB version of the product with a Touch Cover. Microsoft isn't talking about its Surface Pro tablet yet ? that is scheduled to arrive, as previously announced, three months after the initial Surface RT product launch.

On Monday, Microsoft's Windows president Steven Sinofsky and Surface team general manager Panos Panay walked reporters through an extensive demo of the product, along with a behind-the-scenes look at how the product was designed and manufactured.

The latest specs
During the presentation, new specs and information came to light. The device will run the Nvidia Tegra 3 T30 processor?with 2GB of RAM. Both the front and back cameras are 720p, meaning it will be good for video (and Skype video calling), but it won't be very useful for still images other than quickie snapshots (not that many people shoot pictures with tablets ... yet). It has two microphones and stereo speakers, and an HD video out port (adapter sold separately).?

Its "Cover port" is specifically used to dock the powered keyboard cover accessories, but Panay confirmed that in the future, this could be used as a means of connecting interactive accessories. (In case you're wondering about speaker docks and things like that, the best answer at this time is that, because the device has both Bluetooth and Wi-Fi, it supports many audio devices wirelessly, including those that use the DLNA protocol for wireless media streaming.)

Though Microsoft says Surface RT will have a 31.5 watt-hour battery, the company didn't name an exact number of hours it could get. Sinofsky did say it gets "all-day battery life," suggesting 8 or more hours of uptime, and he confirmed that it takes just 2 hours to charge up fully, using the included charger.

How about that touch?keyboard?
Though we only had a brief time to actually use the Surface RT, my first impressions are positive. It's snappy, and the Windows 8 start screen springs to life when you touch it. Flying through a photo gallery is as good as you'd expect, and quite?comparable to an iPad.

What I was really curious about was what it would be like to use the Touch Cover keyboard, and its ease of use came as a pleasant surprise. This isn't some dumb accessory: There's a lot of code written to ensure that the keyboard knows the difference between actual typing and you just setting your fingers down. For instance, if you gently place your finger down on the backspace key, nothing happens. But if you start to press down, it suddenly responds.?

Typing fast will take some practice ??even Panay says the learning curve is four to five days.?When I tried to build up some?speed, I did alright.?The only real issue I had was with the space bar. Though the touchable area of the space bar is pretty large, I couldn't quite manage to hit it, but practice will no doubt improve my space-barring skills.

The other neat feature of the Touch Cover is the integrated track pad. It supports two-finger scrolling, and responds to swipes and taps with problem whatsoever. You quickly start to treat the Surface like a laptop, but given the fact that it comes with a free?full Office 2013?suite ? which is impressive in its own right but isn't particularly touch friendly ? having the laptop-like?mode is a good thing.

Accessories and availability?
If you buy the Surface then want to add accessories later, a Touch Cover by itself costs $119.99, while a Type Cover, the cover with a deeper, mechanical keyboard, costs $129.99. An HD digital AV adapter costs $39.99 and a VGA adapter costs $39.99. Though the product comes with a power supply, you can buy an additional one for $39.99.

Microsoft is adamant that the first wave of Surface tablets is sold through its direct retail channel. You will see it at Microsoft stores around the country ? 27 brick and mortar stores to date in U.S., with four more opening in November, along with 34 more "holiday" kiosk stores popping up in malls all over. You will not see it at any Best Buy or Walmart stores.?

Online shoppers in the U.S. as well as the UK, Canada, Germany, France, Australia, China and Hong Kong will be able to go to Surface.com to buy it as well.

"Is it a tablet or is it a laptop?" Sinofsky asked reporters, rhetorically, in anticipation of the big question most people have. "I've been using this now for a long time, six months, and it's been really fascinating for me," he continued. "I've used a lot of tablets. This is not a tablet, but it is the best tablet I've ever used. And I've used a lot of laptops. This is not a laptop, but it's the very best laptop I've ever used."

We shall see, but from what we did get a chance to experience, this baby is well worth a look come Oct. 26 ? assuming you live close enough to a Microsoft store to get a gander.

Amid ongoing economic crisis, EU celebrates 'Single Market Week'

The EU is celebrating the 20th anniversary of the single market this week, in part to point out that despite the economic crisis wracking Europe, the union has brought positive changes too.

News reports about the European Union nowadays do not give much reason for celebration. Greece is on the brink of economic collapse, other European countries are also in severe economic troubles, and shrinking solidarity is going hand-in-hand with reviving stereotypes. Yes, the European Union won the Nobel Peace Prize last week, but this news was quickly met with cynical comments.

Nevertheless, the EU today kicked off a week of celebrations, commemorating the 20th anniversary of the single market, which allows Europeans and their goods to move around all 27 member states without borders or barriers. The Single Market Week encompasses events all around Europe, with various ?opportunities to discuss the main achievements of the Single Market.? Some of these include ?cheaper calls abroad,? ?safer toys,? and ?more rights for travelers,? according to documents handed out to attendees of the launch event of the Single Market Week at the European Parliament in Brussels.

The celebration comes at a time when citizens' trust in the European Union is at an all-time low, according to the most recent survey by Eurobarometer. Only 31 percent of EU citizens say they trust the European Union (mind you: they trust their national governments even less). Also, 28 percent have a flat-out negative view of the EU ? almost double the figure of six years ago.

According to some of the speakers today, the European Union doesn't always receive the credit it deserves. ?Citizens don't attribute their cheaper mobile phone bills to the single market,? said Malcolm Harbour, a member of the European Parliament for the British Conservative party, during a meeting with journalists. ?Sometimes we forget how much has been achieved. The free movement of people, students studying abroad ? citizens take that for granted.?

Mr. Harbour has a point. I arrived here in Brussels yesterday from the Netherlands, without realizing that my train didn't have to stop at the border. The only reason I brought my passport was to show it at the security check of the European Parliament. And it is not just free movement of journalists and other Europeans. If I want, I can buy as many Belgian waffles as I want without having to declare them at the border.

As European Commissioner Michel Barnier said: ?Our lives have been made easier.? But as with most advancements, we quickly take them for granted. So how to make the people more aware of these things? Mr. Barnier's answer was simple: ?We have to talk about it.? And that's what EU representatives will be doing this week.

Jelly-like atmospheric particles resist chemical aging

ScienceDaily (Oct. 16, 2012) ? Atmospheric chemists at the Harvard School of Engineering and Applied Sciences (SEAS) have found that when it comes to secondary organic material in the atmosphere, there are two distinct breeds: liquids and jellies.

Secondary organic materials (SOM) are airborne particles that have begun to react with gases in the atmosphere. In the last 20 years' research and climate modeling, these SOM particles have been assumed to drift as liquids. In a liquid phase, the organic materials would absorb other compounds like ammonia or ozone very easily and then progress through a series of chemical changes (known as chemical aging) to form particles that reflect or absorb sunlight, or form clouds.

Now, experiments at Harvard, using particles of ?-pinene SOM and adipic acid, have shown that a drop in humidity can send these common aerosols into a jelly-like phase, in which they resist chemical aging almost entirely. The findings, published in the Proceedings of the National Academy of Sciences, may call for a revision of regional and global climate models.

"Our research provides the first experimental evidence that the chemical aging process of atmospheric particles is limited by phase," says principal investigator Scot Martin, Gordon McKay Professor of Environmental Chemistry at SEAS and in Harvard's Department of Earth and Planetary Sciences. "Solid or semi-solid aerosol particles will only react with other molecules at the surface of the droplet, instead of mixing homogeneously. What this means is that the time scale of important chemical aging processes may be much longer than what is reflected in current models."

The two particles chosen for this study, ?-pinene and adipic acid, are common in Earth's atmosphere; ?-pinene is essentially a scent released by coniferous trees (including pines -- hence the name), and adipic acid comes from both anthropogenic sources (such as car exhaust) and natural chemical reactions.

In the atmosphere, the particles of ?-pinene SOM and adipic acid behave rather like gelatin; in moist conditions, the droplets absorb water and remain liquid. In dry conditions, they solidify without crystallizing. Lead author Kuwata Mikinori, a postdoctoral fellow at SEAS, compares these semi-solid atmospheric particles to chunks of tofu, another high-viscosity, amorphous solid.

"If you pour soy sauce onto a block of tofu at room temperature, the liquid will just sit on the surface of the tofu. It won't sink in," Kuwata explains. "But if you cook tofu in a sauce at a high heat for a long time, the tofu will eventually absorb the taste of the sauce. That's the same kind of effect we're seeing in the atmosphere. Eventually these semi-solid aerosols do blend with other reactants, but it takes a long time, a higher temperature, or enough ambient humidity to encourage a phase change back to liquid."

The researchers selected ammonia as the reactant in their study partly because its nitrogen component is easy to detect using mass spectrometry, but also because of its current environmental relevance. Atmospheric ammonia has been on the increase in the past few years as a byproduct of fertilizer use and livestock farming, and as a result of increasing temperatures.

"For the environment, ammonia is a very tricky compound," says Kuwata. "It neutralizes sulfuric acid, helping prevent acid rain, but its nitrogen component can also fertilize open bodies of water, which can be bad news for ecosystems. When ammonia reacts with SOM, it can form ammonium salts, which are thought to affect cloud nucleation activity, and organic nitrogen, which forms light-absorbing compounds."

Martin and Kuwata conducted their experiments in the Harvard Environmental Chamber, a 5-cubic-meter Teflon bag that hangs from the ceiling of an environmentally controlled laboratory room at SEAS. In trials within this chamber, they recreated various atmospheric conditions while adjusting the humidity, and exposed particles of either ?-pinene SOM or adipic acid to ammonia for approximately seven minutes.

Following that exposure, they measured the diameter of the resulting particles and determined the mass and composition of each one to understand the extent of chemical aging that had occurred.

"Our results challenge basic assumptions about the rate of chemical reactions in the atmosphere," says Kuwata. "These results ought to change the way we evaluate the impacts of atmospheric aerosol particles on the climate."

This work was supported by the U.S. Department of Energy and by a postdoctoral fellowship from the Japan Society for the Promotion of Science.
